Proteome Factory utilizes an Applied Biosystems Model 491 Procise N-terminal proteine/peptide amino acid sequencer (Edman sequencer) with an on-line ABI Model 140C PTH Amino Acid Analyzer. The N-terminal degradation method used by the sequencer is derived from the chemical process developed by Pehr Edman.
N-terminal Edman sequencing service for proteins and peptides The N-terminal Edman sequencing service is applied to protein and peptide samples delivered lyophilzed, in solution or bound to PVDF membrane. Depending on sample amount typically 5 to 40 residues can be determined by N-terminal sequencing.
Internal amino acid sequencing service The internal amino acid sequencing service is applied to protein samples delivered lyophilzed, in solution or bound to PVDF membrane. Depending on the requested type of service we digest the protein using different endoproteases and/or chemical protein cleavage. The generated peptides are separated and fractionated using an Agilent 1100 capillary HPLC system with microfraction collector ensuring highest sensitivity. Peptide peaks are applied to N-terminal sequencing with or without additional mass spectrometrical analyses like determination of peptide mass, MSMS or de novo peptide sequencing.
